Enzymatic Digestion Solution Ⅲ

Product Description

This digestion solution has been filtered and sterilized, and it can be directly used for the digestion of cultured cells or some tissues. It features convenience and rapidity. Usually, it takes about one minute at room temperature to digest most adherent cells. Generally, when the cells show obvious slight morphological changes and there is no need to wait until the cells become round at all, the cells can be pipetted off.

Instructions for Use

(1) Digestion of Adherent Cells:


  1. Aspirate the culture medium, and wash the cells once with sterile PBS, Hanks' solution or serum-free culture medium to remove the residual serum.

  2. Add a small amount of trypsin cell digestion solution (containing phenol red), just enough to cover the cells slightly, and place it at room temperature for 30 seconds to 2 minutes. The digestion time varies for different cells.

  3. Observe under a microscope. When the cells contract significantly and obvious morphological changes of the cells can be found at the bottom of the culture vessel by naked eyes, or when the cells can just be pipetted off by gently blowing with a pipette, aspirate the trypsin cell digestion solution (containing phenol red). Add the complete cell culture medium containing serum, pipette off the cells, and then they can be directly used for subsequent experiments.

  4. If it is found that the digestion is insufficient, add the trypsin cell digestion solution (containing phenol red) to redigest. If it is found that the digestion time of the cells is too long and some cells have already detached directly from the bottom of the culture vessel before being pipetted, pipette all the cells down with the trypsin cell culture solution. Centrifuge at 1000 - 2000g for 1 minute to pellet the cells. Try to remove the trypsin cell digestion solution (containing phenol red) as much as possible, then add the complete culture medium containing serum to resuspend the cells, and they can be used for subsequent experiments.


(2) Digestion of Tissues:


The digestion time required for different tissues varies greatly. Usually, it is advisable that the tissues can be fully dispersed after digestion.
