Trypan Blue

Product Description

This product is formulated by leveraging the property that normal and healthy cells are capable of excluding Trypan blue, whereas cells with compromised cell membrane integrity can be stained by it. Stringently speaking, Trypan blue staining is employed to assess the integrity of cell membranes. Conventionally, it is assumed that once the cell membrane integrity is lost, the cell is regarded as deceased.


Subsequent to Trypan blue staining, a relatively accurate quantification of cell viability can be achieved either through direct counting under a microscope or by counting after photographing under a microscope.

Instructions for Use

(1) Cell Collection:


For adherent cells, first digest the cells with trypsin and/or EDTA. For suspension cells, the cells can be collected directly. Centrifuge the collected cells at 1000 - 2000 g for 1 minute, discard the supernatant, and then resuspend the cells in 1 mL or an appropriate volume of cell resuspension solution according to the amount of cells.


(2) Trypan Blue Staining:


Pipette 100 μL of the resuspended cells into a plastic centrifuge tube, add 100 μL of Trypan blue staining solution (2X), gently mix them evenly, and stain for 3 minutes (a staining time of 3 minutes is sufficient, although the staining time can be extended a bit longer, but it should not exceed 10 minutes).


Note: For cells resuspended in cell culture medium, an equal volume of Trypan blue staining solution (2X) can also be directly added for staining.


(3) Counting:


Pipette a small amount of the stained cells and count them using a hemocytometer. Generally, if a relatively precise quantification is required, at least 500 cells should be counted for each cell sample, and both the number of blue cells and the total number of cells should be recorded.


Cell viability = (Total number of cells - Number of blue cells) / Total number of cells × 100%.
